Abstract

Aedes aegypti is an important vector of dengue and other mosquito-borne diseases. Entomopathogenic bacteria from the bamboo rhizosphere may provide a potential biological approach for controlling its larvae. This study aimed to isolate entomopathogenic bacteria that are able to suppress Aedes aegypti mosquito larvae in a laboratory environment. Bamboo rhizosphere soil samples were collected at several selected bamboo rhizosphere locations around Mataram City. Two hundred and fifty grams of soil was collected from a depth of 5-10 cm below the surface and placed into pre-sterilized containers. The soil in the containers was stored in a shaded area before use in the study. The bacteria were isolated by heat treatment the soil mixed with sterile distilled water (10% concentration), then conducted to serial dilution and spreading it on Nutrient Agar media supplemented with Amoxicillin and Streptomycin. The growing colonies of bacteria were then tested for their toxicity on Instar III Aedes aegypti larvae in the laboratory to determine their toxicity. Bacterial isolation from all bamboo rhizosphere locations yielded two entomopathogenic bacterial species, Bacillus thuringiensis and Bacillus sphaericus. B. thuringensis isolated from several locations exhibited toxicity to instar III A. aegypti larvae ranging from 30% to 60%, while B. sphaericus exhibited toxicity ranging from 10% to 30%. The toxicity values demonstrated by these two bacteria were categorized as low to moderate. Differences in toxicity levels between isolates confirm the influence of habitat on the bacteria's biological characteristics. This finding reinforces the role of indigenous microorganisms as an environmentally friendly and sustainable source of vector control.
