Effectiveness of Guava Leaf Extract on Hematological Profile of Nile Tilapia Infected with Vibrio vulnificus

Abstract
Nile tilapia (Oreochromis niloticus) is an important aquaculture commodity in Indonesia, but intensive production is vulnerable to bacterial infections, including Vibrio vulnificus. This study evaluated the effect of guava leaf (Psidium guajava) extract incorporated into commercial feed on the hematological profile of Nile tilapia infected with V. vulnificus. A completely randomized design with four treatments and three replications was used. Treatments consisted of P0 (control), P1 (25 mL/kg feed), P2 (50 mL/kg feed), and P3 (75 mL/kg feed). Fish were reared for 45 days, comprising 30 days of feeding treatment and 15 days post-infection. Blood samples were collected on days 0, 30, and 45. Guava leaf extract significantly affected erythrocyte and leukocyte counts (P<0.05). The highest observed erythrocyte counts were recorded in P1, reaching 3.477 × 10⁶ and 3.583 × 10⁶ cells/mm³ on days 30 and 45, respectively. The highest observed leukocyte count occurred in P3, reaching 10.143 × 10⁴ cells/mm³ on day 45. Lymphocyte proportion increased from 65.0% to 75.8%, whereas monocytes and neutrophils decreased. Basophils remained relatively stable. These findings indicate that guava leaf extract influences the hematological response of Nile tilapia following V. vulnificus infection. However, an optimal dose could not be established from the tested concentrations.
